MIT study links APOE4 gene to faulty brain insulation

Researchers at MIT report that the APOE4 gene variant, the biggest genetic risk factor for late-onset Alzheimer's, suppresses Wnt signalling in oligodendrocytes. These are the brain cells that make myelin, the insulation around neurons.
Professor Li-Huei Tsai and Leyla Akay, a former graduate student, led the study in the peer-reviewed journal Cell Stem Cell. When the team restored Wnt signalling, myelination recovered in human cell cultures and in mice modelling Alzheimer's. They did this by inhibiting a protein called GSK3b. The work used cell cultures and mice only, so it has not been tested in people.
